Duke Nukem Forever sells itself on three things: having been in the works a long time, being the latest installment in a series known for inventive, high-action gameplay, and scantily-clad ladies. Duke Nudem, a promotional Flash game presumably released so as to lock down that name before Ron Jeremy snaps it up, features one out of those three things – presumably to get you hankering for the day when they're finally combined.
Duke Nudem challenges you to beat a series of target-shooting challenges, defeating gun-toting action girls in high-score challenges. Lose and the game commands you to remove an item of clothing (woe betide your fellow net-cafe customers). And while it might seem like the only reason nobody's pointing out the juvenile sexism of this whole affair is for fear of stating the obvious, a combination of high AI and woefully sluggish controls sees the game inadvertently making a pretty positive statement about ladies' proficiency with heavy artillery.

Duke Nudem comes with SFW and NSFW options, but it should go without saying that if you work with, you know, adults, “Safe for Work” may still be a bit of a misnomer. How'd you do?
