Assassin's Creed 2, Splinter Cell: Conviction, Halo 3: ODST. Also, Mass Effect 2. My brother picked it up in February, and it was really cool up until I left the Citadel and had to fight. I hated the combat, it felt so weak and lifeless. I've tried it again since then, but I still hated it. In the latest trailer Bioware said that they're improving the combat, I hope they do; I really wanted to play the first one but I couldn't stand that aspect.
ALSO, it would be sweet if Nintendo announced some new actual games. As we all know, last year's conference was major weaksauce.

Well my number 1 concern for E3 is Metal Gear Solid to see if any new MGS game is in the works.

Broken record, but I'd like to see Alan Wake. Been waiting for that forever. Heavy Rain, ME2, any MMO's for console would be great. Also, the spy rpg game. The name escapes me now.

Sly 4 and Mass Effect 2 videos. Something on the new PSP. And maybe a look at future updates for the 360, in particular updates to the NXE. A teaser on Jade Empire 2 would be cool but doubtful.

Capcom's second mystery game at E3 - I am curious as to see what new action game it is. I want it to be either a new Mega Man or a Legendary Wings revival? Why Legendary Wings? Other than a woman with wings packin' heat, no reason.
Final Fantasy 13 - With those three building-sized billboards poppin' up in LA, I hope the 2010 release date is good.
Lego Rock Band on the DS - just to see how it controls.

But, the thing I most want to see at E3 this year is Borderlands. The new graphics made me interested. Now, I want to see this damn game in motion to make sure I'm not picking up a dud.

Something cool or even acceptable from Nintendo.
New dlc for Left 4 Dead some whole new campaigns would be nice.
I'd love to see nintendo exectute Ravi Drums live on stage. Also tits.

I'll start by getting the obvious ones out of the way. A good 2D, old-fashion Sonic would be fantastic, either Mega Man 9-style or like the good parts of Unleashed. Also, definitely a new Kid Icarus. His part in SSBB seems to make it immanent.
I'd really love to see a remake of an old, practically forgotten franchise that will blow us all away, like what they did with Prince of Persia. My game of choice? Altered Beast. This could really make for a sick game with modern technology.
I also would like to see a sequel announced to Okami. What an amazing first game, and I would love one on a new console THAT ISNT WII.
Oh, and I want 14 Guitar Hero games announced, just because it would be hilarious.
